FOXO4-DRI
FOXO4-DRI (Forkhead box O4 D-Retro-Inverso) is a synthetically modified, D-amino acid peptide antagonist designed to disrupt the interaction between the FOXO4 transcription factor and the p53 tumor suppressor protein. In cellular longevity and senescence research, it is studied as a selective senolytic agent. Laboratory investigations focus on its ability to trigger apoptosis specifically in senescent cells by releasing p53 to translocate to the mitochondria, while leaving non-senescent, healthy cells uninjured.
- Senescent cells: Damaged cells that stop dividing but refuse to die, secreting inflammatory signals known as SASP.
- FOXO4 and p53: FOXO4 locks the p53 protein inside the cell nucleus, preventing natural programmed cell death (apoptosis).
- Animal studies: Laboratory tests on aged mice have shown improvements in hair density, kidney function, and overall physical fitness markers. [1]
- Safety status: Studied strictly in vitro and in animal models; lack of human safety data and potential cancer-pathway interactions require high caution.
